Hi, this is Chad with Munday Scientific. Today we’re going to walk you through how to disassemble and reassemble a Nikon 50i microscope.
Disassembly of the Nikon 50i 55i
We’ve got a standard Nikon 50i here. First, we’ll remove the head. This model has a thumb screw—or a knurled screw—right here. Hold the head securely, loosen the screw, and lift the head off.
For the rest of the microscope, you’ll need a set of Allen wrenches, which you can find at any hardware store. Some microscopes have built-in tools—this one does—so we’ll use the Nikon tool included with the scope.
Next, we’ll remove the nosepiece. The Nikon nosepiece can be a little tricky. If you’re standing in front of the unit, look to the right-hand side. You’ll see a hole for a 2 mm Allen wrench. Loosen that screw, and the nosepiece will drop down off the dovetail mount. Set it aside carefully.
Now we’ll move to the stage. Using the same wrench, locate the front mounting point—insert the wrench and loosen the screw. Some Nikon models may use a knurled screw here instead. Once loosened, the stage will lift off.

Next is the substage condenser. There are various types of condensers; this one has a flip-out lens for working with 1x to 100x objectives. On the side of the scope, there’s a screw that secures the condenser. Use the same 2 mm wrench to loosen it, then slide the condenser out.
Check to see if your microscope has a blue filter installed. This one doesn’t, because we retrofitted it with a Nanodyne LED light source, replacing the original halogen system.
Reassembly of the Nikon 50i 55i
Now we’ll reassemble the microscope, following the same steps in reverse.
Start by reinstalling the condenser. Lower the substage for easier access. This model has a locking pin in the back, so insert the condenser and rotate it until it clicks into place.
Next, reinstall the stage. Carefully align it and hold it securely with both hands while tightening the screw. Once it’s locked in, lower the stage to make room for the nosepiece.
For the nosepiece, Nikon uses a specific alignment system. There’s an indentation that needs to match the groove on the dovetail. Start by inserting it halfway—it won’t go in completely at first. Push upward, then slide it back into position. You’ll know it’s properly seated when the front of the nosepiece is flush with the dovetail. Secure it using the same 2 mm Allen screw.

Finally, reinstall the head. Align the flange with the dovetail mount, slide it into place, and tighten the thumbscrew to secure it.
